The Song of Appalachia follows Amira, a recent college graduate, and geneticist, studying the potential links between music and DNA. On a weekend hike through the mountains, she and her colleagues discover a link between their research, and the mountains themselves. Their impromptu research is disrupted when one of their number goes missing, followed swiftly by a second, and a slew of strange occurrences.
As their numbers dwindle and more dangers reveal themselves, Amira and her friends are forced to reconcile with the dangers that hide in the mountains, as well as those hiding in their blood.
